Study found patients who took saxagliptin ( Onglyza & Kombiglyze XR ) had a 27 percent higher risk of heart failure

Concerns over saxagliptin’s connection with heart failure came after the FDA reviewed the SAVOR study, a randomized controlled clinical trial that found patients who took saxagliptin had a 27 percent higher risk of heart failure than patients who did not take the drug. The study was published in The New England Journal of Medicine in October 2013. Based on the study’s findings, the FDA asked saxagliptin’s maker, AstraZeneca, to investigate a potential link between the type 2 diabetes medication and heart failure. In April 2015, the FDA’s Endocrinologic and Metabolic Drugs Advisory Committee voted 14-1 in favor of the FDA requiring saxagliptin to carry a warning label about the risk of heart failure. The committee member who cast the dissenting vote wanted the drug taken off the market.

Onglyza side effects and Kombiglyze XR side effects can cause dangerous side effects that include:

  • Heart failure
  • Thyroid cancer
  • Pancreatitis (inflammation of the pancreas)
  • Pancreatic cancer
  • Lactic acidosis (a build up of lactic acid in the blood)

Onglyza (saxagliptin) was approved by the U.S. Food and Drug Administration (FDA) in 2009. Kombiglyze XR (saxagliptin and metformin hydrochloride extended-release) was approved in 2010. The two medications are prescribed to people who do not suffer from diabetic ketoacidosis, a condition in which increased ketones are in the blood or urine. Onglyza and Kombiglyze XR are dipeptidyl peptidase-4 (DPP-4) inhibitors, a class of drugs that treat type 2 diabetes by inhibiting an enzyme in the body that reduces insulin production. In August 2015, the FDA warned that DPP-4 inhibitors have been associated with severe and disabling joint pain. The warning came after a review of the FDA’s adverse event reports that showed patients started having joint pain symptoms from 1 day to years after they started taking a DPP-4 inhibitor. The FDA added a new warning and precaution about the joint pain risk to the labels of all DPP-4 medicines, including Onglyza and Kombiglyze XR.
